Analisis In Silico Primer dan Probe untuk Deteksi Aspergillus fumigatus dengan PCR

Abstract


Aspergillus fumigatus merupakan patogen utama penyebab aspergillosis invasif. Deteksi infeksi A. fumigatus untuk diagnostik klinik rutin dapat dilakukan menggunakan metode real time Polymerase Chain Reaction (real time PCR). Pemilihan primer dan probe yang tepat merupakan komponen penting untuk digunakan dalam deteksi A. fumigatus dengan metode real time PCR. Studi ini bertujuan untuk memberikan alternatif pemilihan primer dan probe yang tepat untuk deteksi A. fumigatus dengan PCR. Primer dan probe yang telah digunakan dalam beberapa penelitian dianalisis menggunakan Primer BLAST NCBI dan program PerlPrimer 1.1.21. Hasil analisis secara in silico beberapa pasang primer dan probe dengan target gen ribosomal memiliki beberapa komposisi basa berulang dan target spesies yang tidak spesifik. Pasangan primer dan probe yang diprediksi memiliki sensitivitas dan spesifisitas paling tinggi untuk mendeteksi A. fumigatus dari sampel klinis di laboratorium yaitu pasangan primer gen pengkode haemolysin.
